چکیده

The results of lymphocyte functional studies performed with the peripheral blood mononuclear cells (MNCs) of patient Wa with a non-Sezary T-ceIl leukemia are described. The aggressive clinical course and the distinctive histologic, cytochemical, and cytogenetic features of this T-cell lymphoproliferative disorder are detailed in an accompanying report. Immunologic marker analysis revealed that 76% of the Wa MNC fraction formed rosettes with sheep erythrocytes (E-rosettes), and on histologic examination, 95% of the E-rosettes contained neoplastic lymphocytes; 10% of the MNCs formed EAC rosettes; and 1 3% showed SIg positivity by immunofluorescence. In vitro mitogenic responses of Wa cells measured by 3H-thymidine incorporation were extremely low. and responses to candida and streptococcal antigens were absent. Although a normal percentage of B cells was evident in the Wa MNC fraction, no Ig synthesis occurred in either pokeweed mitogen (PWM) stimulated or unstimulated cultures. In coculture experiments with control MNCs from two normal individuals, the Wa cells exhibited no suppressor or helper effects on the normal PWMstimulated lg synthesis. Importantly however, the Wa cells demonstrated suppressor effects on the normal mixed lymphocyte reaction (MLR) occurring between populations of allogeneic MNCs from two normal donors. The degree of MIR suppression increased with the presence of

Monoclonal antibody analysis of human T lymphocyte subpopulations exhibiting autologous mixed lymphocyte reaction.

In autologous mixed lymphocyte reaction (MLR), T cells proliferate in response to the stimulation by autologous non-T cells. In the present study, human T cell subpopulations, defined by murine monoclonal antibodies OKT4, OKT8, or 9.3, were examined for their capacity to proliferate in autologous and allogeneic MLR. It was observed that the treatment of responder T cells with OKT4 or 9.3 antibo...

Induction of suppressor activity in the autologous mixed lymphocyte reaction and in cultures with concanavalin A.

T lymphocytes that are activated in the autologous mixed lymphocyte reaction (MLR) have suppressor activity. Concanavalin A (Con A) augments the suppressor activity generated in cultures containing both T and non-T lymphocytes and can induce suppressor activity in T-lymphocyte preparations that contain too few (10%) non-T cells to generate a significant autologous MLR. However, when such T-lymp...

Regulatory mechanisms in cell-mediated immune responses. IV. Expression of a receptor for mixed lymphocyte reaction suppressor factor on activated T lymphocytes

Suppression of the mixed lymphocyte reaction (MLR) by a soluble factor produced by alloantigen-activated spleen cells requires genetic homology between the factor-producing cells and responder cells in MLR. The ability of lymphocytes used as MLR responder cells to adsorb MLR suppressor factor was tested to investigate the expression of a receptor structure for suppressor molecules. Normal splee...

Effects of anti-interleukin-2 receptor antibody and cyclosporin A on human T cell proliferation in primary mixed lymphocyte reaction.

Effects of anti-IL2 receptor antibody (anti-IL2 RAb) and cyclosporin A (CsA) on human T cell proliferation in primary mixed lymphocyte reaction (MLR) were investigated. Both agents inhibited the proliferative response induced by alloantigen in a dose dependent manner when they were added at the initiation of culture. We also analyzed the expression of activation antigen on responder cells and t...
